【摘要】 探索和建立稳定的犊牛口腔黏膜上皮细胞体外培养模型。采取新生犊牛口腔黏膜组织,中性蛋白酶和胰酶消化后,将获得的口腔黏膜上皮细胞接种于含10%胎牛血清的培养基中连续培养,探索最适培养条件,对细胞进行形态学观察和免疫组织化学染色,并用电镜观察其超微结构。结果表明,体外培养的细胞为单一的上皮细胞,无成纤维细胞混杂,细胞角蛋白免疫组化染色阳性,透射电镜观察可见细胞间特有的桥粒结构。说明体外连续培养犊牛口腔黏膜上皮细胞获得成功,为口腔黏膜体外研究提供了实验模型。更多还原

【Abstract】 To explore and establish a model of culture of calf oral epithelial cells.Specimens from calf were dissociated into single cell suspension by dispase and trypsin.The cells were grown in a medium with 10% fetal bovine serum and explore the best qualification of cells growing.The cells were grown in vitro to observe morphological characteristics.The results showed that the cultured cells were all epithelial cells without fibroblasts.The observation of electron microscope revealed that desmosomes between the cells,and the immunohistochemistry of cytokeratin antibody showed positive staining.The study showed that the culture technique for growing oral epithelial cells from calf has been established and provided an experimental model for further study of oral mucosa.更多还原
