基因芯片筛选肉鸡胫骨软骨发育不良差异表达基因

【摘要】 为研究肉鸡胫骨软骨发育不良发病机理,应用基因芯片技术筛选福美双诱导的胫骨软骨发育不良早期的差异表达基因.提取对照组和饲喂福美双组第1、2、6天的生长板总RNA,分别与18张基因芯片杂交,采用real-time PCR对7个基因进行差异表达验证.结果表明,基因芯片筛选到第1、2、6天1.5倍以上差异表达基因数分别为108、385和468.赖氨酸氧化酶(LOX)、热体克蛋白25(Hsp25)、DNA结合1抑制剂(ID1)、动联蛋白1(KTN1)、分泌性卷曲相关蛋白(SFRP4)、钙黏蛋白1(CDH1)和烯醇酶2(EN02)等7个基因上调表达与芯片结果一致。为认识肉鸡TD的病因、从分子水平阐明其发病机制提供了可靠的生物信息学依据.
【Abstract】 The aim of this study was to select the genes differentially expressed in the growth plate of broiler chickens with tibial dyschondroplasia(TD)induced by thiram with gene chip.RNA was extracted from the growth plates of control and thiram-fed Chickens at d 1,2 and 6.Then they were hybridized with eighteen gene chips respectively,and differential expression genes were also identified by real-time PCR.The results showed that the data identified 880 differentially expressed probe sets,which corresponds to 108,385,and 468 probe sets exhibiting 1.5-fold changes at d 1,2 and 6,respectively.The expression of lysyl oxidase(LOX),heat shock protein 25(Hsp25),inhibitor of DNA binding 1(ID1),kinectin 1(KTN1),secreted frizzled-related protein 4(SFRP4),cadherin 1(CDH1)and enolase 2(ENO2)were up-regulated,which corresponded to the results by real-time PCR.Our findings may provide reliable bioinformatics basis on understanding the molecular pathogenesis of TD.
