高氟对雏鸡肝细胞周期和凋亡影响的研究

【摘要】 选用1日龄Avian肉鸡健雏300只,随机分为4组,分别饲以对照日粮(F 23mg/kg)和高氟日粮(F 400mg/kg,Ⅰ组;F 800mg/kg,Ⅱ组;F 1200mg/kg,Ⅲ组)42天,以流式细胞术的方法研究了高氟对雏鸡肝细胞周期和凋亡的影响.高氟Ⅱ组和高氟Ⅲ组肝脏绝对重量显著低于对照组(P<0.05),脏器指数显著高于对照组(P<0.05).流式细胞仪测定,高氟Ⅰ组各期肝细胞比例和凋亡率均无显著变化.14日龄时高氟Ⅱ组和高氟Ⅲ组肝细胞G0/G1期极显著升高(P<0.01),S期、G2+M期和增值指数(PI)与对照组相比不同程度地降低;28和42日龄时高氟Ⅱ组和高氟Ⅲ组肝细胞G0/G1极显著降低(P<0.01),S期和PI值显著或极显著升高(P<0.05或P<0.01),G2+M期无显著变化.14、28和42日龄时,高氟Ⅱ和Ⅲ组肝细胞同时凋亡率均极显著高于对照组(P<0.01).结果表明,日粮含氟800mg/kg及1200mg/kg时可引起肝细胞增殖能力增强,凋亡细胞显著增多.

【Abstract】 The experiment was conducted with the objective of examining the effect of dietary high fluorine on the cell cycle and apoptosis of liver in chickens by the methods of flow cytometry(FCM).300 one-day-old Avian broilers were divided into four groups and fed on Control diet(F 23.00mg/kg)and High fluorine diets(F 400.00mg/kg,High fluorine groupⅠ;F 800.00mg/kg,High fluorine groupⅡ;F 1200.00mg/kg,High fluorine groupⅢ)for 6 weeks.The result showed that the weight of liver were significantly decreased and the relative weight of liver were significantly increased in high fluorine groupⅡand high fluorine groupⅢthan in control group.By FCM,there were no marked changes on the cell cycle and apoptosis-rates of liver in high fluorine groupⅠ.The G0/G1 phase was significantly increased,and S phase,G2+M phase and PI(Proliferating index) decreased in diversity in high fluorine groupsⅡandⅢat 14 days of age.The G0/G1 phase was significantly decreased,S phase and PI significantly increased,and G2+M phase was not changed in high fluorine groupⅡand high fluorine groupⅢwhen compared with those of control group at 28 and 42 days of age.The apoptotic rate was higher in high fluorine groupⅡand high fluorine groupⅢthan in control group at 14,28 and 42 days of age.It was concluded that fluorine 800mg/kg diet and 1200mg/kg diet could cause enhanced multiplication capacity of hepatocyte and increased apoptotic.
