中药成分复方及其组分药对新城疫苗免疫应答的影响

【摘要】 为探讨中药成分复方及其组分药对鸡免疫功能的影响,将饲养至14日龄的非免疫健康罗曼蛋公鸡594羽随机均分为11组,除空白对照组外均用NDV-Ⅳ系疫苗滴鼻、点眼免疫,28日龄二免。在首次免疫的同时,1~9组分别口服高、中、低剂量药物,疫苗对照组和空白对照组口服等量生理盐水,每天1次,连续3天。分别于14、21、28、35和42日龄采血,用MTT法测定T淋巴细胞增殖的动态变化,用β-微量法测定血清抗体效价动态变化。分别于14、28和42日龄,每组随机抽取4羽翼静脉采血,分离血清,用双抗夹心ELISA法测定血清中IL-2浓度。结果表明,中药成分复方能增强雏鸡对新城疫疫苗的免疫应答,促进T淋巴细胞转化,显著提高血清抗体效价,明显促进血清中IL-2产生。中药成分复方中、高剂量的效果较好,优于其单味组分药。

【Abstract】 The study was designed to investigate the effects of cCHMIs and their components on immune function in Chicken.Five hundred and ninety-four 14-day-old non-immunized Roman cocks were randomly divided into 11 groups.Groups 1 to 9 were OM-APS,OM and APS at high dose,middle dose,and low dose, respectively.Group 10 and 11 were vaccine control and blank control,respectively.Groups 1 to 10 were inoculated with NDV-Ⅳvaccine by intraocular-nasal route,repeated vaccination at 28 days old.At the same time of the first vaccination,the chickens in groups 1 to 9 were given compounds at high,middle and low doses,respectively,by oral administration,but in vaccination group and blank control group with physiological saline,once a day for 3 days.On days 14,21,28,35 and 42,peripheral T lymphocyte proliferations and serum antibody titers were determined by MTT method andβ-microscopic method,respectively.On 14,28 and 42 days,the IL-2 concentration in the serum was determined by ELISA method.The results showed that cCHMIs can enhance chickens immunologic response to ND vaccine,which promoting the transformation of T lymphocytes,improving antibody titer and increasing IL-2 concentration in the serum.cCHMIs at high and middle doses were obviously higher than single Chinese herb.
