LTB基因对奶牛乳腺炎核酸疫苗的作用评价

【摘要】 目的检测LTB基因在乳腺炎核酸疫苗中是否可起到免疫增强的效果。方法制备两种质粒pcDNA3.1-FnbpA和pcDNA3.1-LTB-FnbpA,经鼻黏膜免疫小鼠,用间接ELISA法检测抗体水平,及进行攻毒比较。结果含有LTB基因的实验组抗体水平显著高于对照组(P<0.05),攻毒结果显示其保护率也显著高于对照组(P<0.05),保护率增强了15.64%。结论 LTB基因可增强乳腺炎核酸疫苗的免疫效果,可以作为乳腺炎核酸疫苗的黏膜免疫佐剂。
【Abstract】 [Objective]The objective of this test was to establish the effect of the LTB gene on vaccine against the dairy mastitis.[Method]Two kinds of DNA plasmids of pcDNA3.1-FnbpA and pcDNA3.1-LTB-FnbpA were constructed,and vaccined the mice mucosa.The antibody was tested with methods of ELISA,and the mice with or without vaccine were invatided by staphylococcus aureuse.[Results]The antibody level of the mice vaccine with the pcDNA3.1-LTB-FnbpA was significantly higher than that of the mice vaccine with the pcDNA3.1-FnbpA(P<0.05),and the mice vaccined against staphylococcus aureuse displayed the same the result with an 15.64%increace of the protection.[Conlusion]The LTB gene enhanced the mastitis DNA vaccine protecting effect,and could be a kind of mucosal DNA vaccine immunological adjuvant.
