奶牛乳腺炎金黄色葡萄球菌溶血型及溶血素基因分布的研究

【摘要】 目的:建立金黄色葡萄球菌溶血型分型及hla基因、hlb基因分型的方法,初步研究生鲜牛乳中金黄色葡萄球菌溶血素基因的分布状况,并探讨溶血型与溶血素基因分型的相关性。方法:利用表型生化分型,利用PCR分型技术对其进行分型,电泳检测扩增结果。结果:以分离自牛生鲜乳来源金黄色葡萄球菌为研究对象,表型分型56株表现为α溶血,检出率为43.4%;43株表现为β溶血,检出率为34.11%;另有29株表现为不溶血,检出率为22.48%;其中hla基因检出率34.88%,hlb基因检出率为42.60%。结论:在溶血表型中,α溶血要多于β溶血;hlb基因的检出率比hla基因的检出率要高。金黄色葡萄球菌所检出溶血素基因与血琼脂平板出现的溶血现象并非一一对应关系。此调查可为金黄色葡萄球菌引起的奶牛乳腺炎的防治提供依据。

【Abstract】 Object:To develop a typing method for Staphylococcal hemolysin genes,and to research on the distribution of hemolysin phenotype and hemolysin genes in Staphylococcus aureus isolates from raw milk of cows and to analysis there relevance.Methods:The hemolysin phenotype was oberserved with the method of blood plate.Hemolysin genes were amplified by PCR,and detected by electrophoresis.Results:The typing method for Staphylococcal hemolysin phenotype and genes with PCR was developed.The isolates withα- hemolysis was 56,account for 43.4%;43 showed beta-hemolysis, account for 34.11%;Another 29 isolates showed no hemolysis phenotype,and it account for 22.48%.The isolates with hla gene account for 34.88%,hlb gene account for 42.60%.Discussion:In the hemolytic phenotype,α- hemolysis are more than beta-hemolysis;however the hlb gene detection rates is higher than hla gene.The distribution of hemolysin phenotype and hemolysin genes is not a corresponding relationship of Staphylococcus aureus isolates from raw milk of cows.The study can provide a basis for prevention and treatment of mastitis in dairy cows caused by Staphylococcus aureus.
