【摘要】 猪是乙型脑炎病毒重要的储存和越冬宿主,在人乙型脑炎流行中扮演着重要的作用。在本研究中,利用RT-PCR技术从2009年山西运城某猪场疑似乙型脑炎仔猪脑组织扩增出乙型脑炎病毒prM和E基因,并通过BHK-21细胞传代,分离到一株病毒。间接免疫荧光和RT-PCR进一步证实该病毒为猪源乙型脑炎病毒,并命名为SX09S-01。根据prM基因绘制的进化树表明SX09S-01毒株属于基因Ⅰ型,E基因与SA-14、SA-14-14-2、P3、HW和XJ69株的核苷酸同源性分别为88%、87%、88%、88%和98%,其蛋白具有强毒株典型特征。这些结果表明从山西运城新分离的乙型脑炎病毒SX09S-01株属于基因Ⅰ型猪源乙脑病毒,为进一步研究猪乙型脑炎病毒流行病学和分子生物学奠定基础。更多还原

【Abstract】 Swine is an important reservoir and overwinter host of Japanese encephalitis virus and plays a critical role in the human epidemic encephahtis.In this study,the prM and E fragments were amplified by RT-PCR from brains of piglets which were suspected to be infected with Japanese encephahtis virus in Yunchen,Shanxi province 2009.A virus was isolated by passage in BHK-21 cells with brain tissues.The isolated virus was confirmed to belong to JEV by immunofluorescence and RT-PCR and named SX09S-01.The isolate belongs to genotypeⅠby the phylogenetic analysis based on prM gene.There were the homogeneousness of 88%,87%,88%,88%and 98%between the E gene of SA-14,SA-14-14-2,P3,HW,XJ69 and SX09S-01.E protein had some typical characteristics of strong virulent JEV.These results confirmed that the SX09S-01 belonged to the genotypeⅠJEV originated from swine,and provided an important basis for further study of JEV epidemiology and molecular biology.更多还原
