【摘要】 通过接种鸡胚成纤维细胞、聚合酶链式反应(PCR)技术及特异性单抗的间接荧光抗体反应(IFA),从某大型肉用型种鸡场的疑似J亚群白血病的病鸡中,以及25个临床健康的商品代肉鸡群的2群中,分离鉴定出J亚群禽白血病病毒(ALV-J)。在用抗ALV-Jgp85单克隆抗体JE9的IFA中,来自病鸡群的两株病毒SD9901和SD9902呈强阳性反应,来自临床健康肉鸡群的YZ9901和YZ9902株呈弱阳性反应。对YZ9901株和SD9902株的PCR产物直接测序结果表明,这二个中国株的囊膜蛋白gp85的氨基酸组成与原型株HPRS-103及美国分离株ADOL2Hcl有89%～93%的同源性,它们相互间的同源性是92%。其3c端LTR区与原型株HPRS-103则有95%～97%的同源性,但中国的两株病毒在紧靠3c端LTR的/E0成分中,出现了一个139个碱基的缺失性突变,而代之以一个11个碱基的插入序列。更多还原

【Abstract】 Subgroup J avian leukosis viruses(AL V2J ) were isolated f rom two broiler breeder farms wit h suspect2 ed diseased chickens and two commercial broiler flocks wit hout clinical symptoms by inoculating t he samples into chicken embryo fibroblast cells and PCR amplification of t he infected CEF genomic DNA.In t he indirect fluo2 rescence assay(IFA) wit h AL V2J specific monoclonal antibody J E9,2 st rains,SD9901 and SD9902 f rom breed2 er wit h suspected lesions,were st rongly positive,anot her 2 st rains,YZ9901 and YZ9902 f rom commercial broilers wit hout clinical symptoms gave weak reactions.The genomes of st rains YZ9901 and SD9902 were par2 tially sequenced and t he result s indicated t hat t heir gp85 had 89%-93%identity in t he amino acid level wit h AL V2J protot ype HPRS -103 and American st rain ADOL2HCl.The amino acid identit y among t hemselves was 92%.The 3 ’noncoding L TR region had 95%-97%identit y in t he nucleotide level wit h AL V2J protot ype L TR region and got an insertion of 112base f ragment instead.更多还原
