不同日粮蛋白水平对猪脂肪组织脂类合成代谢相关基因表达的影响
Impact of Dietary Protein Levels on Lipid anabolism Related Gene Expression in Adipose Tissue of Wujin Pigs

【摘要】 本研究探讨不同日粮蛋白水平对乌金猪脂肪组织脂类合成代谢相关基因表达的影响。选取健康、体重约15kg的乌金猪27头,随机分为3组,设3个蛋白水平,在15~30kg,30~60kg和60~100kg生长阶段分别为低蛋白水平(14%、12%和10%)、中蛋白水平(16%、14%和12%)和高蛋白水平(18%、16%和14%)。乌金猪体重为30kg、60kg和100kg时屠宰取皮下脂肪组织样品,提取总RNA,反转录,Real-time PCR定量检测脂肪组织脂肪合成代谢相关酶和因子的基因表达水平。结果表明:高蛋白日粮显著降低乌金猪脂肪组织脂肪合成代谢相关基因(ACC、FAS和SREBP-1c)在60kg和100kg体重时的表达,而30kg体重时FAS mRNA被显著提高。该结果指示,高蛋白日粮可显著影响脂肪组织脂类合成代谢相关基因及脂肪酸转运基因在不同体重时的基因表达。
【Abstract】 The experiments were performed to identify the molecular mechanism that different protein levels on fat deposition in animal adipose tissue.[Method]Twenty-seven Wujin pigs were randomly assigned to 3 groups and imposed on three different protein levels with low dietary protein(14%,12%and 10%),middle dietary protein(16%,14%and 12%) or high dietary protein(18%,16%and 14%) during the growth stages of 15~30,30~60 and 60~100kg body weights.Pigs were slaughtered and subcutaneous adipose tissue were taken for analyzing lipid anabolism related gene expression by real-time PCR.The results showed that:High dietary protein could significantly reduce the gene expression of ACC,FAS and SREBP-1c at the body weights of 60kg and 100kg,but the mRNA concentration of FAS was increased at 30 kg body weight.The mechanism for different dietary protein levels to impact adipose tissue lipid filling is through regulating the gene expression of enzymes and factors for fatty acid synthesis and transport.
