ChM-Ⅰ在肉鸡胫骨软骨发育不良的时序性研究

【摘要】 (目的)本实验旨在研究ChM-Ⅰ在肉鸡胫骨软骨发育不良(Tibial Dyschondroplasia,TD)中不同阶段的表达情况,进一步对肉鸡胫骨软骨发育不良发病机理进行探讨。(方法)150羽一日龄艾维因内鸡饲喂1周后分为两组,对照组饲以基础日粮,试验组饲以基础日粮中添加100mg/kg的福美双,2d后试验组改换饲喂基础日粮,历时20d。应用自行制备的兔抗ChM-Ⅰ多体对饲喂福美双后的第1、2、4、6、8、10、12、15d的肉鸡胫骨软骨生长板进行免疫组化研究。(结果)结果显示肥大区的ChM-Ⅰ在第1d和第10d对照组的表达量要高于试验组,在第2、4、6、8、12、15d对照组的表达量要低于试验组。(结论)结果表明,ChM-Ⅰ在不同时期的表达有差异,但总体趋势表明试验组ChM-Ⅰ的表达量还是高于正常组的表达量,由此可推测ChM-Ⅰ可能在TD发生过程中起到重要作用,可能是生长板无法血管化和钙化的重要原因。

【Abstract】 (objective) This experiment is aim at study the expression of ChM-I in tibial dyschondroplasia (TD) of broiler in the different stages,and further to explore the pathogenesis of the broiler tibial dyschondroplasia.(method)150 one-day AA broiler chickens were randomly divided into two groups after being raised seven days.the control group was given a regular diet,whereas the experimental group was given same diet containing thiram 100mg/kg for two days,then both of the groups were given the same regular diet,the feeding experiment lasted 20 days.Immunohistochemistry was applied to detect the synthesis of ChM-I..at the first day,the second day,the forth day,the sixth day,the eighty day,the tenth day,the twelfth day,the fifteenth day after raised thiram,and the self-maded rabbit anti-ChM-I IGg was used for the firt anti-body.The resμlts expressed that the control’s quantity of ChM-I in the hypertrophic zone was more than.experiment ’at the firth day and the tenth day,but the control’s quantity of ChM-I in the hypertrophic zone was less than.experiment ’at the second day,the forth day,the sixth day,the eighty day,the twelfth day and the fifteenth day,so the experiment ’squantity of ChM-I was more than control’from the tendency of overall.we can conduct that ChM-I play an important role in tibia growth plate that can’t vascularized and calcified.
