铜导致肉鸡肝细胞游离钙超载作用的研究

【摘要】 铜是动物机体的必需微量元素之一,对生命体的正常生长发育起到关键性作用。为进一步了解铜对生物体的生物学效应,我们从亚细胞结构层次设计了本次实验。选用硫酸铜作为实验铜源,向即时分离的肉鸡肝细胞悬液中加入不同剂量的铜(铜终浓度分别为:0μmol/L、5μmol/L、10μmol/L、20μmol/L、30μmol/L),孵育不同时间(孵育时间分别为:5、10、15、20、30分钟)后观察其细胞内游离钙浓度的变化。结果显示:高剂量的铜可以破坏细胞内游离钙稳态,钙内流造成细胞内钙超载,铜浓度20μmol/L和30μmol/L刺激肝细胞30分钟后,其[Ca2+]i显著高于对照组(0μmol/L)(p<0.05)。肝细胞在10μmol/L Cu2+环境中孵育不同时间后显示,时间越长,细胞内钙浓度越高。表明,铜(Ⅱ)能诱导细胞内游离钙的超载,并具有浓度依赖性和时间依赖性,暗示铜对体细胞的毒害作用可能是通过改变细胞内游离钙浓度来实现。
【Abstract】 Copper is an essential trace element in animal body,it play a pivotal role on the lives of normal growth and development.In order to further learning about its biologic function,this research was carried out to understand the effect of copper on intracellular free calcium in broiler hepatocyte.After incubating the broiler hepatocyte with copper(from copper sulfate(Ⅱ)) of different concentration(the final concentration of Cu2+ were 0μmol/L,5μmol/L,10μmol/L,20μmol/L,30μmol/L respectively) for different time(incubation time were 5,10,15,20,30 min respectively),the changes of its intracellular free calcium were observed.In results,exposure to a high dose of copper for 30 min can leads to disruption of [Ca2+]i homeostasis and result in overload of intracellular free calcium in broiler hepatocyte,the[Ca2+]i in 20μmol/L and 30μmol/L group is much higher than control group(p<0.05);When the broiler hepatocyte exposured to 10μmol/L Cu2+ for different time,the results manifested that the longer incubation time,the higher of[Ca2+]i.The results obtained showed that copper(Ⅱ) can induce overload of[Ca2+]i in broiler hepatocyte and these effects had dose-dependent and time-dependent which indicated that the poisoning effect of copper on somatic cells maybe through the way of inducing overload of intracellular free calcium.
