【摘要】 35日龄100羽伊沙蛋鸡随机均分为对照组(含钙1.00%)和高钙组(含钙3.78%)。试验第17天,对两组鸡作血清中尿酸、钙、磷、肌酐和尿素氮的测定,并各组剖检5只鸡,作肾脏的光镜和电镜观察,并用原位缺口末端标记法(TUNEL法)对肾小管细胞凋亡细胞进行染色和计数,计算凋亡细胞百分率。结果表明,与对照组相比,高钙组血清无机磷显著降低,血清钙、尿酸、肌酐和尿素氮显著升高。光镜观察,高钙组肾小球内有大量尿酸盐,有的肾小球代偿性肥大;肾小管上皮细胞肿胀,出现颗粒变性,部分肾小管上皮细胞坏死(与基膜脱高),有的肾小管管腔变小,甚至闭锁;远曲小管管腔内有嗜中性粒细胞。电镜观察,高钙组近曲小管上皮细胞出现核固缩,部分细胞溶解、坏死、微绒毛脱落,线粒体肿胀、部分空泡化,近曲小管基部初面内质网扩张;间质纤维母细胞轻度增生,淋巴细胞漫润;肾小球毛细血管袢触突微绒毛化;肾小囊足细胞轻度溶解、水肿,部分线粒体空泡化。与对照组相比,高钙组肾小管凋亡细胞百分率显著升高。更多还原

【Abstract】 One hundred healthy growers of 35 days of old were randomly divided into two groups.The growers in different groups were fed with normal calcium diet(control,1.00%calcium),high calcium diet (HC,3.78%calcium) respectively.At the end of experiment,we detected electrolyte of serum.5 growers in each group were dissected,and kidney sampled and observed by optical microscope and electronmicroscope.TdT-mediated dUTP-biotin nick-end labeling(TUNEL) assay was used to detect apoptosis of renal tubular cells.The results showed serum urine acid,calcium,creatinine and urea N increased in the HC group,meanwhile,Serum phosphorus decreased in the HC group as compared with the control.Microscopical examination showed that Lots of urate deposited in glomeruli of HC a grower.The epithelial cells of renal tubule swollen,degenerated and putrescence.The antrum of dital convolute tubule have lots of neutrophil.Electonmicroscopical observation found that the nucleus of proximal tubule contracted.Rough-surfaced endoplasmic reticulum of proximal tubule of a HC grower expanded.The cells of fibre ventre was hyperplasia and lythphocyte sokaged.The cells of fibre ventre was hyperplasia and lythphocyte sokaged.The sticking of glomeruli capillary vessel were chorioned.The cells of renal theca dissolved and dropsied,and revealed an expanded mitochondrion with vacuoles.The rate of apoptosis of renal tubular cells increased in the HC group as compared with the control.In conclusion,high calcium diet could cause renal injury and increase the apoptosis of renal tubular cells in growers.更多还原
