中国2006-2007年狐、貉和水貂源犬瘟热病毒基因分型和H基因遗传多样性分析

【摘要】 为研究近年来在我国狐、貉和水貂中流行的犬瘟热病毒(Canine distemper virus,CDV)基因分型和变异情况,本研究通过RT-PCR扩增了来自2006~2007年间我国不同省份共13株狐、貉和水貂源CDV野毒株H基因全长,并对其进行克隆测序。通过与GenBank上已发表的另外6株CDV野毒H基因氨基酸序列比对分析发现,除HL株与其它CDV野毒株氨基酸同源性较低外(92.3%-94.1%),另外18株之间氨基酸同源性较高(96.1%-99.5%),而包括CDV3和Onderstepoort株等弱毒疫苗株的H基因氨基酸序列与以上19株CDV野毒同源性均较低(90.1%-91.4%)。对以上19株CDV野毒基因分型结果显示,其分别归属Asia-Ⅰ和Arctic型2个基因型,其中18株属于我国CDV流行的主要基因型——Asia-Ⅰ型,与我国犬源代表株TN株和guizhou株等处于同一基因型,仅有HL株属于Arctic基因型。研究结果表明,当前在狐、貉和水貂中流行的CDV野毒株与国内所用的疫苗毒株CDV3株的H基因(主要保护性抗原基因)序列存在较大的差异,推测近期某些地区CD的流行和免疫失败现象的发生可能与CDV野毒株H基因变异导致抗原性的改变有关。

【Abstract】 To characterize the genotypes and genetic variation of haemagglutinin(H) gene of canine distemper virus(CDV) from foxes,raccoon dogs and minks in China,H gene of 13 strains of CDV from different hosts and geographical areas in China between 2006 and 2007 were amplified by RT-PCR and then sequenced.The deduced amino acid sequences of the H protein were compared with those of other 6 strains of wild-type CDV from the GenBank,It showed all the 19 strains wild-type CDV have a high homology(96.1%-99.5%) with each other except for the HL strain.While they all have a low homology(90.1%-91.4%) with vaccine strains of Onderstepoort and CDV3.Phylogenetic analysis of the H gene of the 19 strains CDV show 18 strains relegated to genotype Asia-I and a narrow clustering closed to the previous field isolates from infected dogs and other animals in China,While the HL strain corresponded to Arctic genotype.So the far heredity between vaccine strain and the wild-type strains of CDV may be responsible for the prevalence of CD in several provinces of China.
