新型鸭肝炎病毒的分离鉴定

【摘要】 从近年来在广东地区流行的鸭肝炎病例中分离到1株病毒(命名为"GD株")。对分离的病毒进行了鸭胚传代培养。DE2-DE6鸭胚毒的ELD50在10-5.17-10-6.38/0.2mL之间。鸭胚毒以100 ELD50/0.2mL接种可以致死80%以上的10-11日龄易感鸭胚,接种鸭胚原代肝细胞单层可以产生细胞病变。DE4代毒对鸭胚的ELD50与对雏鸭的LD50基本一致,回归雏鸭后可以产生与1型鸭肝炎病毒相同的临床症状和解剖病变。鸭肝组织毒对4、8和16日龄雏鸭的LD50分别为10-6.5/0.5mL、10-5.38/0.5mL和10-4.83/0.5ml。病毒对氯仿、乙醚和胰酶不敏感,能够耐受pH3.0酸处理,62℃30分钟不能被完全灭活。病毒核酸类型鉴定为RNA病毒。生物学和理化学特性鉴定结果表明,GD株为强毒鸭肝炎病毒。使用标准1型DHV阳性血清进行的交叉中和试验和交叉被动保护试验表明,GD株病毒在抗原性上与1型鸭肝炎病毒无关,属于一种在我国流行的新型鸭肝炎病毒强毒(暂命名为"CN-DHV")。

【Abstract】 A virus strain(designated GD) was isolated from death ducklings with duck hepatitis occured in Guangdong recently. The isolated virus was passed with duck embryos.The virus titers of DE2~DE6 passages adapted to duck embryos are from 105.17to 106.38 ELD50(50%embryos lethal dose) per 0.2 ml.The duck embryos viruses can cause the death of over 80%duck embryos with 10-11 days-old by inocubation with 100 ELD50 per 0.2 ml and the cell-pathogenic effects of primary duck embryos liver cells.The DE4 virus contains the ELD50 to duck embryos similar to the LD50(50%lethal dose) to ducklings.The intramuscular injection of duck embryos viruses to ducklings can cause the same clinical symptoms and pathological changes caused by the type 1 duck hepatitis virus(DHV-1).The LD50 of duck liver tissue virus to the 4-,8-and 16-days-old ducklings are 10-6.5,10-5.38and 10-4.83 per 0.5 ml,respectively.The virus is not sensitive to chloroform,diethyl ether and trypsin and possesses acid-resisting and heat-resisting features.The GD strain belongs to RNA virus showed by nueic acid type test.The biological and physicochemical characterization revealed that GD strain is a virulent DHV.The GD strain was determined to be antigenically unrelated to DHV-1 by in vitro cross-neutralization assay and in vivo cross-protection test with standard antiserum against DHV-1.Therefore,GD strain is a new serotype of DHV occurred in China(designated CN-DHV).
