大鼠前扣带回和海马在非快速眼动睡眠中对记忆的选择性再启动

【摘要】 哺乳动物睡眠期间非快速眼动睡眠和快速眼动睡眠交替出现,行为学和电生理学研究都表明睡眠具有记忆巩固作用。非快速眼动睡眠参与记忆巩固的形式主要有两种:"自发性再启动"和"线索性再启动"。已有证据表明在非快速眼动睡眠期间,大鼠海马对先前学习的空间记忆痕迹不但有自发性再启动,而且此期间重新接受学习过程中的气味刺激可强化空间记忆。有研究记录大鼠前扣带回的神经元放电活动,结果表明:与任务活动相关的特征性放电序列在随后两小时内的非快速眼动睡眠中再启动。本实验的研究目的:探索前扣带回和海马在非快速眼动睡眠中参与线索性再启动的神经活动特点,以及线索性再启动所对应的具体学习内容。应用多通道神经元放电同步记录技术,观察大鼠在学习前后的非快速眼动睡眠呈现学习相关气味线索时的神经活动,以及学习线索性空间记忆过程中两脑区相应的放电特征。结果发现:前扣带回和海马存在特征性神经元,它们在学习前后的非快速眼动睡眠中对气味线索的放电频率显著增高;通过对两脑区神经元放电序列时间分布的相似性分析,发现前扣带回和海马共同参与线索性再启动过程。结果提示奖赏相关的线索性空间记忆需要前扣带回和海马的共同参与,再启动过程同样需要二者的协同合作。
【Abstract】 Non-rapid eye movement sleep and rapid eye movement sleep alternating present during mammal’s sleep.Recently,Behavioral and electrophysiological studies have shown that sleep has a role in memory consolidation non-rapid eye movement sleep involved in memory consolidation,there have been two main forms:spontaneous restart and cued restart.There have been evidences that during the non-rapid eye movement sleep of rats, Spatial memory is provided with spontaneous restart,moreove,emergeing of Learning-related odor cues will lead to memory consolidation.And researchers have recorded rat anterior cingulate cortex neuronal firings,which indicate that there has been a restart of the spike train in task-related neurons in the subsequent 2 hours.Purpose of the present study:research characteristics of neural activity participation clued restart in hippocampus and anterior cingulate cortex,and specific learning content corresponding to clued restar.With the multichannel single-unit recording technique,we observe the neural activities and the firing patterns in anterior cingulate cortex and hippocampus in non-rapid eye movement sleep before and after the acquirement of the rules when learning-related odor cues appear.The results found there are specific neuron in anterior cingulate cortex and hippocampus,and a significant increase in firing rates in non-rapid eye movement sleep of specific neurons during the appearance of the cues. And also,with the analysis of the similarity of temporal distribution of spike train,it proves the cooperative processing of anterior cingulate cortex and hippocampus during the restart.The result suggests reward-related cues of spatial memory requires participation of both anterior cingulate cortex and hippocampus and collaboration of these two regions is required for restart.
