四川松茸遗传多样性
Genetic diversity of Tricholoma matsutake in Sichuan

【摘要】 为了了解四川松茸的遗传资源及其多样性,以采自四川小金、雅江、木里等7个县的24株不同生境的野生松茸子实体为材料,采用ISSR(Inter-Simple Sequence Repeat)标记及ITS测序分析了这些菌株的遗传多样性。从15条ISSR引物中筛选出2条多态性较好的引物扩增所得条带表明,ISSR标记显示,在松茸中存在一定的遗传多样性;亲缘关系树状图表明,来自木里、会东、小金的大部分松茸样品聚为一个类群,而雅江、盐源、盐边、冕宁的为另一类群,出现了一定地理分化。ITS序列分析表明,松茸菌株间的ITS序列相似性很高。同时系统进化树表明四川松茸Tricholoma matsutake与美洲口蘑Tricholoma magnivelare同源关系较近,与欧洲口蘑Tricholoma caligatum距离较远,这与报道的形态学分类并不一致。本研究表明四川具有丰富的松茸遗传资源。
【Abstract】 In order to understand the genetic resources and diversity of Tricholoma matsutake in Sichuan Province,twenty-four wild fruit bodies of T.matsutake were collected from different sites in 7 counties such as Xiaojin,Yajiang,Muli and so on.The genetic diversity of these strains was revealed by ITS sequencing and ISSR analysis.Polymorphic bands were generated with 2 primers chosed from 15 primers indicated that certain diversity was conserved in these strains.The dendrogram of genetic relationships demonstrated that most of the strains collected from Muli,Huidong and Xiaojin were clustered together,while most the strains collected from Yajiang,Yanyuan,Yanbian and Mianning were clustered into another group,indicating the geogrphical isolation of T.matsutake in Sichuan.The T.matsutake collected from different sites showed highly homologous.In addition,the T.matsutake in Sichuan showed highly homologous with Tricholoma magnivelare but lowly with Tricholoma caligatum in the analysis of phylogenetic tree,which was not consistent with morphological classification.In conclusion,the resources of T.matsutake in Sichuan was abundant.
