草菇菌丝细胞核迁移研究

【摘要】 草菇菌丝细胞为多核细胞,每个细胞内的细胞核数目不定、大小不一,且存在能移动的迁移核。本研究通过对草菇菌株V23、T1、T2、T3、T4、T5、T6、T7、T8、T9、T10、T11、T12、T13、T14、T1 5的菌丝进行结晶紫染色,并对其菌丝细胞进行显微观察,其结果显示:处于生长中的菌丝,其顶端的细胞通常无核或少核,而菌丝中间的细胞核数目较多;除T2、T4、T1 5菌株外,其它菌株的菌丝均存在明显的迁移核,且有时能观察到中心体;所有能判断其移动方向的迁移核,其迁移方向均指向菌丝顶端;在细胞质中,迁移核呈蝌蚪状,在穿越细胞间通道时,迁移核有呈蝌蚪状、哑铃状、细丝状等,没有固定形状。动力蛋白的动力输出是细胞内小泡运输、有丝分裂时的核迁移和纺锤体移动等一系列生命活动的关键动力;动力蛋白激活蛋白可激活动力蛋白,也可促使动力蛋白与运输货物结合。本研究克隆草菇动力蛋白激活蛋白基因的部分片段,采用半定量RT-PCR对其表达进行分析,其结果显示:T2、T4、T15菌株的表达量明显比其它菌株的表达量低。这些结果表明:草菇菌丝细胞中,细胞核并非只是随细胞质的流动而移动,存在主动迁移,迁移核具有向菌丝顶端迁移的特性,中心体是其动力源,而动力蛋白可能是其分子马达,受动力蛋白激活蛋白调控。

【Abstract】 Vegetative hyphae cell of Volvariella volvacea is a multinucleate cell.The number and the size of the nucleus are unsteady in each cell and migrating nuclei can move in vegetative hyphae.Nuclei of vegetative hyphae were stained by eosin crystal violet staining for strains V23,T1,T2,T3,T4,T5,T6,T7,T8,T9,T10,T11, T12,T13,T14 and T15,and then their hyphae cells were observed by microscope in this study.The results showed there was no nucleus or few nuclei in top cells of hyphae and more nuclei in middle cells of hyphae when the hyphae were growing rapidly.Except for strains T2,T4 and T15,migrating nuclei were found in all others,and sometimes centrosomes were observed in vegetative hyphae.The migratory direction pointed to the top of hyphae in all of migrating nuclei whose migratory direction could be judged.Migrating nuclei were tadpole-shaped in cytoplasm and had no definite shape,such as tadpole-shaped,dumbbell-shaped and filiform-shaped,when the migrating nuclei were passing through intercellular channel.The key powers of a series of life activities come from the output power of dynein,such as vesicular transport,migration of nucleus and spindle in mitosis.Dynactin can activate dynein or prompt dynein combine with cargo.The part fragment of dynactin was cloned and its expression was analyzed by semi-quantitative RT-PCR in this paper.The results showed that the expression level of strains T2,T4 and T15 were lower than that of other strains.The results indicated that nuclei were not just moved with the flow of cytoplasm and existed in the active migration.In vegetative hyphae,migratling nuclei have the character of migration towards the top.The power source comes from centrosome and dynein that is regulated by dynactin may be molecule motor.
