红斑革孔菌化学成分及其生物活性研究

【摘要】 红斑革孔菌(Coriolopsis sanguinaria(Kl.)Teng)为多孔菌科(Polyporaceae),革孔菌属(Coriolopsis)的真菌,生于腐木上,常引起木材白色腐朽。目前尚未见红斑革孔菌活性次生代谢产物的研究报道。前期研究结果表明其发酵提取物具有一定的生物活性,为此,本研究采用液体发酵对红斑革孔菌进行发酵培养,并分别对其菌丝体和发酵液化学成分进行研究。采用硅胶柱层析法、凝胶柱层析法、重结晶等多种方法对发酵产物的乙酸乙酯提取浓缩液浸膏进行逐级分离纯化,通过物理常数法、化学方法和波谱法[核磁共振(NMR)和质谱(MS)]等对化合物进行结构鉴定。目前已分离获得10个化合物,鉴定出5个化合物,其中化合物1为酪醇,化合物2为diaporthein A,化合物3为麦角甾醇,化合物4为啤酒甾醇,化合物5为过氧化麦角甾醇。化合物1和2为首次从该属真菌中分得,酪醇具有抗氧化作用,diaporthein A具有一定的抗结核分枝杆菌(Mycobacterium tuberculosis)活性,MIC为200 mg/mL。
【Abstract】 Oosporein,a mycotoxin,has been isolated from the fungi of the genera Beauvearia, Chaetomium,Phlebia,Verticillium and Aphanocladium.As a toxin,oosporein may cause kidney injury of poult,and even be fatal under high dietary level.But on the other hand,oosporein has significant insecticidal and antiviral activities,and may have applications in the control of insect pests and the cure of plant diseases.Actually,oosporein is relatively safe for it can hardly be adsorbed by plant and(or) can be rapidly detoxified by plant,so it is rarely detected in organism and environment. In this paper,the entomogenous fungi of Cordyceps cardinalis is firstly reported to be able to produce oosporein in submerged cultures.Oosporein is abundant in fermentation broth and can be easily extracted.Its antibacterial and antitumor activities are also reported here. Extraction and purification of oosporein.The strain C033 of Cordyceps cardinalis was fermented in C8 medium,special for cultivation of Cordyceps spp.The fermentation broth was separated from mycelium by centrifuging,and then was extracted by ethyl acetate.The extract liquor was concentrated to obtain the crude extraction.The crude extraction was treated by"alkali-solution and acid-precipitation"method(dissolved in 1 M NaOH,and precipitated by adding 98-99% HCl) to make a precipitation.The precipitate was processed in the same way three times,and then washed by water,dried and weighted.The product is a red pigment material,with yield 0.12 g/L. Structure determination of oosporein.The purity of the red pigment material was examined by HPLC,and the result shows that its purity reaches 99.3%.The NMR and MS were used to determine its structure,and this red pigment material is confirmed as oosporein. Testing the activities of oosporein.The antitumor and antimicrobial activities were tested by MTT method and 96-well plate assay method respectively.The results show that it has inhibitory rates of 30.9%against Candida albicans and 65%against Breast Cancer Cell MCF-7 at a low concentration of 50μg/ml,displaying moderate antibacterial and antitumor activities. In summary,Cordyceps cardinalis can be used as producing strain of oosporein.Together with other oosporein production fungi,Cordyceps cardinalis can be used as original strain in genome shuffling,which is demonstrated as an effective new approach for strain improvement,to obtain high yield strains with industrial production prospect of oosporein.
