【摘要】 麦角甾醇又称麦角固醇,是真菌类的特征甾醇,广泛存在于药用真菌子实体和发酵菌丝中,是真菌菌丝细胞膜的构成物质基础(Deng et al.2001)。茶藨子叶状层菌Phylloporia ribis(Schumach.:Fr.) Ryvarden为锈革孔菌科真菌,是忍冬根茎部生长的真菌子实体,具有抗炎、抑菌、抗肿瘤等作用(Zhang & Dai 2005;Dai & Yang 2008)。现代研究认为,药用抗癌真菌主要通过免疫调节作用来实现其对癌症的治疗作用,即其有效成分能调动或活化机体的"内源性抗癌物质",包括活性细胞和活性因子,使机体处于平衡、稳定与和谐的状态,防止或拮抗肿瘤的发生与发展(Mizuno T 2002;Kang & Chen 2005)。药用真菌中普遍存在的麦角甾醇及其过氧化物可抑制胆固醇生物合成酶的活性(Deng et al.2001),具有抑制化学物质诱发的肿瘤生成、抗炎、抗氧化等重要的药理作用。更多还原

【Abstract】 Ergosterol namely ergosterin is a characteristic sterol of fungi,widely exists in medicine fungi fruit body and fermented hypha,is also a basic constituent of hypha cell membrane. Phylloporia ribis(Schumach.:Fr.) Ryvarden belongs to Hymenochaetaceae,grows in root and rhizome of Lonicera japonica Thunb.,it has the activities of anti-inflammation,anti-bacterial and anti-tumor.Studies showed that the anti-tumor fungi acted through immunoregulation.Its active components stimulate the inner induced sources including active cells and actors to make the body to stay in balance,stable and harmony status,and inhibit the genesis and evolution of tumor.The widely presented ergosterol and its peroxides in medicine fungi are inhibitors of cholesterol biosynthetic enzyme,they can inhibit the tumor occur,anti-inflammation, anti-oxydation. To use the medicine fungi resource,we fermented the hypha of the species and established a ergosterol quantitative determination method by RP-HPLC to controll the quality of fermented Phylloporia ribis hypha extracted by petroleum ether.Agilent 1100 Zorbax eclipse XDB C8, (150mm×4.6mm) column was used with methanol as mobile phase.The flow rate was 1.0 ml/min and detected at 282 nm.The calibration curve was liner within the range of 0.1～0.5mg/ml.The limitation of quantitation was 0.1mg/ml.The average relative deviations(RSD) was 0.52%.The method of RP-HPLC determination of free ergosterol in Phylloporia ribis (Schumach.:Fr.) Ryvarden is a simple,sensitive and accurate.更多还原
