一株产磷脂酶细菌的筛选及其性质研究

【摘要】 采用卵黄平板法从江苏省兴化市沙沟村等油菜地采集土样中分离到16株磷脂酶A1产生菌株,通过摇瓶复筛得到1株磷脂酶A1产量较高细菌。生长特性及产酶特性研究结果表明:该菌株最适生长及产酶温度均为30℃;最适生长和最适产酶pH值分别为7.0和8.0;最适生长及产酶装液量都为20%;最适生长和产酶碳源为卵黄液、氮源是NH4NO3。菌株所产磷脂酶A1最适作用温度为50℃,最适反应时间为20min,最适大豆卵磷脂底物浓度为0.4%。
【Abstract】 16 Phospholipase A 1 -producing strains were isolated from soil of the rape land in Shagou village, Xinghua city, Jiangsu province by the methods of egg yolk Plate. Among of them, one bacteria strain which produced the higher phospholipase A 1 was obtained through second screening in shake flasks. The results of characteristics on growing and enzyme-production showed that the temperature of its optimum growing and phospholipase production conditions were all 30℃, the pH value of optimum growing and phospholipase production conditions was 7.0 and 8.0 respectively. Its optimum medium volume were 20%.Its optimum carbon source and nitrogen source of growing and phospholipase production were egg yolk and NH 4 NO 3 . The optimum acting temperature and time were 50 ℃ and 20 minof phospholipase A 1 produced by screening bacteria. The optimum soy lecithin substrate concentration was 0.4%.
