高效液相色谱法测定竹茹提取物及其不同部位中的腺苷含量

【摘要】 建立一种反相高效液相色谱(RP-HPLC)法测定竹茹提取物及其不同部位的腺苷含量的方法。采用RP-HPLC法;Luna C18ODS柱(4.6 mm×250mm,5μm),乙腈-0.01 mol/L磷酸缓冲溶液(pH=6.5);体积流量:1.0 mL/min;进样量:10μL;柱温:40℃;检测波长:257 nm。结果采用RP-HPLC法竹茹提取物及其不同部位的腺苷含量,在0.121.124μg/mL范围内线性关系良好(R2=0.9999),平均回收率为98.1-99.1%,RSD为1.98-3.11%,检测限为40 ng/mL。本方法简便、灵敏、准确,可普遍应用于竹茹提取物的质量研究和品质控制。

【Abstract】 To develop a method for determination of adenosine in extract of bamboo shavings by RP-HPLC.RP-HPLC method was used with Luna C18ODS(4.6 ×250mm,5 μm) column.The mobile phase was acetonitrile-0.01 mol/L phosphate buffer solution(pH =6.5) with 1.0 mL/min flow rate;The column temperature was 40 ℃;The detection wavelength was set at 257 nm.RP-HPLC method was used for the determination adenosine;The calibration curve of adenosine was linear under the content 0.12-124 μg/mL;The linearity the average recovery was from 98.1-99.1%,.RSD was 1.98-3.11%,limite of detection is 40 ng/mL.Conclusion The method is simple,sensitive and accurate.The method can be adapted to control the quality of extract of bamboo shavings.
