基于银纳米颗粒聚集致拉曼光高增强的等离子体ELISA超灵敏莱克多巴胺检测技术
Surface Enhanced Raman Scattering-Based Plasmonic ELISA for the Ultrasensitive Detection of Ractopamine

【摘要】 为了能更好地满足对食品添加剂、农药兽药残留等有害物质高灵敏检测的需求,本实验在传统的ELISA检测技术上,利用局域表面等离子共振和表面增强拉曼光谱的高灵敏信号,开发出新的ELISA信号检出系统,可实现待测物的超灵敏检测。该方法中银纳米颗粒的适度聚集可高增强拉曼标记物4MBA的拉曼信号,过氧化氢可氧化聚集的银纳米颗粒从而使拉曼信号下降,酶联免疫吸附试验(ELISA)中的标记的酶可消耗过氧化氢。这样,检测物的浓度通过酶联免疫反应中酶的变化与拉曼信号的变化连锁起来。我们以瘦肉精莱克多巴胺为检测对象,可以在和ELISA相近的时间和步骤内完成检测,检测灵敏度达到1fg/ml。该方法不但灵敏度高、简便快速,而且信号检出系统可适用于各种已成熟的ELISA的检测,具有通用性。本技术仅通过改变ELISA的底物信号系统就可成数量级提高检测的灵敏度,对于有超低含量检测需要或严禁检出的食品污染物,具有很好的应用前景。
【Abstract】 For the requirements of high sensitive detecting to hazardous substances,such as food additives and pesticide and veterinary drug,here,we demonstrate a novel method to use Raman signal as signal-generation system of ELISA,which combines SERS with plasmonic ELISA(SERS-pELISA),for ultra-sensitive analyte detection in ELISA,We present a simple Raman signal-enhanced mechanism,the aggregation of silver nanoparticles generate more SERS hot-spot.The enzyme label of an ELISA controls the dissolve of Raman reports-labeled silver nanoparticles through H2O2 and generates Raman signal when the analyte is present.These methods make SERS-pELISA be able to achieve ultra-sensitive detection under similar operation and time as conventional ELISA.Ractopamine were detected in urine at the ultralow concentration of 1fg/ml.This new method possesses not only ultra-sensitivity and simpleness,but also robustness because of its characteristic.It has a good application prospect for detecting to hazardous substances,especially the hazardous substances that is forbidden to check out.
