免疫彩色二氧化硅纳米微球新型凝集技术快速检测鸡白痢鸡伤寒沙门氏菌
A New Agglutination Test Based on the Immune Colored Silica Nanopartides for Rapid Detection of Salmonella pullorum and Salmonella gallinarum

【摘要】 目的:探寻鸡白痢鸡伤寒沙门氏茼快速检测的新型凝集技术。方法:本研究尝试用3种有机活性染料与抗体分别修饰二氧化硅活性微球制成免疫彩色二氧化硅微球(Immune colored silica nanopatticles,ICSN),构建快速检测鸡白痢鸡伤寒沙门茵新型凝集技术。利用反相微乳法制备彩色二氧化硅纳米微球(Colored silica nanoparticles,CSN),用扫描电镜表征形貌特征和分散程度,通过凝集现象表征对鸡白痢鸡伤寒沙门氏茵的检测效果。结果:该新型凝集技术灵敏度好,其凝集结果醒目、直观,易于肉眼判别,且耗用抗体不到常规凝集试验的1/500,对目标茵的检测范围为10~2~10~9 cfu/mL;重复性和稳定性好,4℃放置28 d后,凝集效果与放置前无显著差异;具有较好的特异性和准确性。结论:用ICSN构建的新型凝集技术具有灵敏、经济、稳定、简便、快速、准确等优点,不仅适用于快速检测鸡白痢鸡伤寒沙门氏茵,还可为其它致病微生物快速检测提供研究基础模型。
【Abstract】 The aim of this study is to propose a new agglutination test for rapid detection of Salmonella pullorum and Salmonella gallinarum(S.pullorum and S.gallinarum).Silica nanoparticles ware modified with three kinds of organic reactive dye and antibody to prepare the immune colored silica nanoparticles(ICSNs),which as the carrier of the novel agglutination test was proposed for rapid detection S.pullorum and S.gallinarum.The colored silica nanoparticles(CSN) were synthesized by reverse microemulsion method.The morphology and degree of dispersion of CSN were characterized by SEM.The detection effect of S.pullorum and S.gallinarum by the agglutination test was characterized.The results showed that the new agglutination test was very sensitive,the aggregation results were prominently,intuitive and easy to distinguish by naked eye,moreover,consumption of antibody in new agglutination test is only 1/500 of that consumed in traditional test,the linear range for S.pullorum and S.gallinarum was from 102 to 109 cfu · ml-1.It was very stable and repeatable,after being stored at 4 ℃ for 28 days,the aggregation effect showed no significant difference;It also showed good specificity and accuracy.(Conclusion) The new agglutination test was simple,fast,accurate,sensitive and economic.Using the novel agglutination test not only detects rapidly S.pullorum and S.gallinarum,but also provides a basis model for rapid detection of other pathogenic bacterium.
