壳聚糖-脂质双分子层的相互作用对脂质体稳定性的影响

【摘要】 基于生物大分子表面修饰的脂质体作为一种新型的输送载体已逐步应用于保健品和功能食品领域。本实验旨在从膜脂流动性和结构的角度研究不同构象壳聚糖分子的修饰对脂质体稳定性的影响。运用荧光各向异性和电子自旋共振技术考察壳聚糖与脂质双分子层的相互作用。结果发现,壳聚糖的舒展分子链和卷曲分子链不仅能通过电荷作用吸附于脂质体表面形成物理屏障,并且对脂质分子的有序性堆积有明显的固化作用。而壳聚糖线圈通过疏水相互作用渗透脂质体双分子层疏水核心,破坏了脂质分子的有序堆积,增大了膜的流动性。尤其在高于临界聚集浓度时,壳聚糖具有高分子表明活性剂的典型特征。此外,显微图像发现,壳聚糖与磷脂分子的相互作用不仅影响脂质体的超微结构和体系分散性,而且对脂质囊泡的聚集、融合也有显著的调控作用。
【Abstract】 The nanocapsules based on biopolymer-decorated liposomes have become an attractive delivery system in nutraceuticals and functional foods.This study was conducted to elucidate the contrasting modulation of chitosan conformation in the stability liposomes.Lipid bilayer-chitosan interactions were investigated by fluorescence anisotropy and electron paramagnetic resonance.It was demonstrated that,in addition to providing the physical barrier for membrane surface,the decoration of chitosan extended and crimpled chains rigidified the lipid membrane.However,the presence of chitosan coils disturbed the membrane organization due to their shallow penetration into the hydrophobic interior of bilayer.Microscopic observations revealed that chitosan adsorption not only influenced the original morphology,size and size distribution of liposomes,but also modulated the particle aggregation and fusion.Especially the role of chitosan coils as a "polymeric surfactant" was confirmed.
