乳铁蛋白脂质体模拟婴儿胃肠道消化的结构和不规则碎片信息图

【摘要】 采用薄膜分散法制备乳铁蛋白脂质体,研究其在成人和婴儿体外胃肠道消化过程中的理化及微观结构的变化。以平均粒径,粒径分布,电位,微观结构,乳铁蛋白的分解,脂质的分解作为消化过程不同时间的评价指标;采用动静态光散射技术研究脂质体消化过程中分形维数(df、ds)随胆固醇及蛋白质的变化规律,进而阐释脂质体消化过程的不规则碎片信息图。DLVO理论用于解释脂质体消化过程中胶体的聚集与絮凝。结果表明,乳铁蛋白脂质体受胃蛋白酶的影响较小,成人和婴儿无显著差异;乳铁蛋白脂质体在肠道中明显破坏,且成人比婴儿变化更显著。脂质体的分形维数受胆固醇及蛋白质影响。相反,流体力学半径(Rh)在不同条件脂质体消化过程中趋于稳定。这些结果为合理设计在不同人群体内具有控释功能的脂质体运载体系提供技术指导,同时为开发新型功能食品控释系统,提高食品营养素的利用率作为指导。

【Abstract】 lactoferrin-loaded liposomes were prepared using a thin-layer dispersion method,the aim was to study the process of change in their physical,chemical and microscopic structures in vitro digestion in the gastrointestinal tract in adults and infants.The average size,size distribution,zeta potential,microstructure,proteolysis of interfacial proteins and lipolysis of the emulsion droplets were monitored as a function of digestion time;static and dynamic light scattering techniques used liposomes digestive process research fractal dimension(df,ds) variation with cholesterol and protein,and thus explained the digestion process liposome fractal hologram.DLVO theory would be the tool to explain the digestion process liposome aggregation and flocculation of colloids.The results demonstrated that lactoferrin-loaded liposomes were little influenced in SGF containing pepsin,adults and infants are no significant difference;lactoferrin-loaded liposomes significantly damaged in simulated intestinal fluid(SIF),and more significant changed in infants than adults.The fractal dimension of liposomes affected by cholesterol and protein.On the contrary,hydrodynamic radius(Rh) was found to be stable during digestion.These results for the rational design in different populations in vivo release of liposome delivery systems have functions to provide technical guidance,as well as the development of new functional food release system,improve the utilization of food nutrients as a guide.
