胡柚不同极性组分对3T3-L1脂肪细胞摄取葡萄糖的影响及其降血糖成分分布的研究

【摘要】 目的:将胡柚果肉分离成石油醚组分(LP)、二氯甲烷组分(MD)、乙酸乙酯组分(EA)、正丁醇组分(NA)、水组分(WF)五个组分,研究这五个组分单独和联合作用对3T3-L1脂肪细胞摄取葡萄糖的影响,并测定胡柚中具有降血糖功效成分的分布及含量。方法:以诱导成熟的3T3-L1脂肪细胞为模型,5个组分分别加入微量的乙醇提取物(EE1),该组分中主成分的含量几乎未变,但却增加了其他4个组分的成分,以此来研究组分间联合作用的降血糖效果,并测定类黄酮、类胡萝卜素、柠檬苦素的分布及含量。结果:除正丁醇组分外,其他4个组分加入EE1后,联合作用与单组分作用相比,可显著促进3T3-L1脂肪细胞摄取葡萄糖;石油醚组分(LP20+EE1、LP30+EE1)、乙酸乙酯组分(EA20+EE1、EA30+EE1)和水组分(WF10+EE1、WF20+EE1、WF30+EE1)与胡柚乙醇提取物的联合作用低于它们单独作用效果之和,二氯甲烷组分和正丁醇组分与胡柚乙醇提取物的联合作用效果与它们单独作用效果之和相近;胡柚中降血糖成分黄酮类化合物主要分布在乙酸乙酯组分中,柠檬苦素类化合物主要存在于二氯甲烷组分中,β-胡萝卜素主要存在于石油醚组分中。结论:(1)胡柚果肉的5个不同极性组分均能促进3T3-L1脂肪细胞摄取葡萄糖。(2)石油醚组分、乙酸乙酯组分和水组分与胡柚乙醇提取物促进3T3-L1脂肪细胞葡萄糖消耗的效果的联合作用低于它们单独作用效果之和,表明其中有某些物质会发生相互拮抗作用。

【Abstract】 The Citrus changshan-huyou pulp was separated five fractions with different polarities,these five fractions all could increase the glucose uptake in 3T3-L1 adipocytes.Based on this research,we study which components combined can increase the glucose uptake in 3T3-L1 adipocytes,and detect the distribution and content of flavonoids,carotenoids,and limonin compounds.Method:The differentiation of 3T3-L1 cells was induced,In order to contrast,add 1 mg/L ethanol extracts(EE1) to ethanol extracts,composition proportion were unchanged,respectively,add EE1 to five fractions,components in other 4 fractions were increased,Based on this theory,study the effects of combination between components,and detect the distribution and content of flavonoids,carotenoids,and limonin compounds.Results:In addition to n-butyl alcohol fraction,other 4 components with trace ethanol extracts,the effect of promoting the glucose uptake in 3T3-L1 adipocytes were increased The Light petroleum fraction(LP20 + EE1,LP30 + EE1)ethyl acetate fraction(EA20 + EE1,EA30 + EE1) and water fraction(WF10 + EE1,WF20 + EE1,WF30 + EE1) promote the effect of 3T3-L1 adipocytes absorb glucose is lower than the sum of their separate effects.The methylene dichloride fraction and n-butyl alcohol fraction combined with ethanol extracts promote the effect of 3T3-L1 adipocytes absorb glucose is close to the sum of their separate effects.Flavonoids are mainly distributed in the ethyl acetate fraction,limonin compounds mainly exist in methylene dichloride fraction,β-carotenoid mainly exist in Light petroleum fraction.Conclusion:(1) Five fractions all can promote 3T3-L1 adipocytes absorb glucose.(2) The Light petroleum fraction、 ethyl acetate fraction and water fraction combined with ethanol extracts promote the effect of 3T3-L1 adipocytes absorb glucose is lower than the sum of their separate effects,and the effect is very significant.Some components happened antagonism effect.
