【摘要】 本实验旨在建立柱前衍生高效液相色谱法测定牡蛎中牛磺酸含量的方法。实验中采用水煮法提取牡蛎中的牛磺酸（提取温度为100℃,提取时间为1h）,分别用HCl和NaOH溶液除去其中的酸性蛋白质和碱性蛋白质,经浓缩、乙醇脱水后制得牛磺酸样品,用邻苯二甲醛（OPA）柱前衍生3min。用乙腈-超纯水（20:80,V/V）为流动相在Agilent Eclipse XDB-C₁₈（4.6×150mm,5μm）色谱柱上分离,流动相流速为0.50mL/min,检测波长360nm,柱温为30℃。牛磺酸和其它成分完全分离,牛磺酸在5μg/mL～60μg/mL范围内线性关系良好（R²=0.9982）,最低检出限0.01957μg/mL;平均回收率为96.97%（RSD=1.97%）,精密度、稳定性、重现性良好（RSD<5%）。该方法专属性强,灵敏度高,定量准确,可用于精确测定牡蛎中的牛磺酸含量。更多还原

【Abstract】 The aim of this paper is to establish a HPLC method for the determination of taurine in oyster with the aid of pre-column derivation technique.The taurine sample was obtained by the combined procedure consisting of extracting the oyster with water at 100℃for 1 h,removing the acid and alkaline protein by the treatment of HCl and NaOH,concentration by the vacuum rotary method,dehydration by the treatment of ethanol and pre-column derivation by the reaction with orthophthalaldehyde（OPA）for 3 min. Pre-treated taurine sample was HPLC separated on a Eclipse XDB-C₁₈ column（4.6×150mm,5μm）eluted with an mixed mobile phase in composition of 20%acetonitrile and 80%ultrapure water at 0.50 mL/min and 30℃.The signal was detected by a UV detector at 360 nm.The taurine showed complete separation from other impurity in the sample.Meanwhile,the method displayed good linearity in the concentration range of 5μLg/mL～60μg/mL with an R² of 0.9982,low detection limit at 0.01957μg/mL,sufficient recovery at 96.97%and desirable relative standard deviation（RSD）of 1.97%for analysis the purified taurine.In addition,The method gave high precision,good stability and repeatability for the determination of taurin sample of oyster with a RSD<5%.The proposed method is accurate,specific and sensitive to determine taurine in oyster.更多还原
