海洋蛋白肽和核苷酸配伍剂对小鼠的免疫调节作用及其机制的研究

【摘要】 目的:探讨海洋蛋白肽(MPP)和核苷酸(NT)配伍剂对小鼠的免疫调节作用及其可能的机制。方法:以小鼠为动物模型,观察不同剂量(0.795g/kgBW、0.265 g/kgBW和0.088 g/kgBW)的MPP和NT配伍剂对小鼠体重、免疫器官相对重量、细胞免疫功能、体液免疫功能、单核巨噬细胞功能、NK细胞活性、血清免疫球蛋白水平以及脾脏淋巴细胞亚群的影响结果:与对照组比较,配伍剂对脾脏脏体比、ConA诱导的小鼠淋巴细胞增殖能力、迟发型变态反应(DTH)程度、抗体生成细胞功能(IgM-PFC)、血清免疫球蛋白(IgG、IgM和IgA)水平、睥淋巴细胞中CD4~+T辅助细胞(Th细胞)百分比以及CD4~+/CD8~+的比值均有显着提高;对脾淋巴细胞中CD4~+CD25~+T调节细胞(Tr细胞)百分比有显着降低;且低剂量组(0.088g/kgBW)效果最好各剂量组对小鼠体重增长、胸腺脏体比、血清半数溶血值(HC50)、单核-巨噬细胞吞噬功能以及NK细胞活性均无影响。结论:MPP和NT配伍剂具有增强小鼠免疫功能的作用,可能通过增强Th细胞功能、促进体内抗体生成以及抑制Tr细胞功能而实现的
【Abstract】 Objective To observe the effects of Marine Protein Peptide(MPP)combined with nucleotides (NT)on immunnomodulation in mice and the possible mechanism.Methods Female Balb/c mice(6~8 weeks old,n = 160 Were administered the(MPP + NT )for 4 weeks with the dose of 0.795g/kgBW,0.265 g/kgBW and 0.088 g/kgBW.Spleen and thymus were weighted and cell - mediated immune functions,humoral immune functions,phagocytic functions of mononuclear phagocyte,NK cell activity assays,the level of immunoglobulin in serum and the splenocyte subpopulations by the flow cytometer were exzamed.Results In comparison with the control group.(MPP+NT )could increase the ratio of spleen and body weight, the capacity of lymphocyte proliferation induced by ConA,DTH response,IgM - PFC number,the level of serum(IgG,IgM and IgA)and the percentage of CD4~+T helper(Th)cell in spleen;and reduce the percentage of CD4~+CD25~+ T regular(Tr)cell.Furthermore,the lowest dose group(0.088g/kgBW )had the most obvious effect,and the difference had statistical significance.Nevertheless,no obvious effects on weight increasing, the ratio of thymus and body weight,the level of serum HC50,phagocytosis capacity and the activity of NK cells were observed in our study.Conclusion MPP combined with NT could improve the immune functions in mice,and may be by the mechanism of enhancing the function of Th cells and the secretion of antibodies and inhibiting the function of Tr cells.
