杜香挥发油抑菌机制研究

【摘要】 目的:杜香是一种生长在长白山的杜鹃花科中草药植物,本实验通过研究杜香挥发油(EOs)对大肠杆菌细胞膜、分泌蛋白表达量及基因组DNA表达量的影响,旨在探讨天然产物杜香挥发油对革兰氏阴性大肠杆菌的作用机制。方法:采用紫外可见分光光度分析法、荧光光谱扫描、SDS-page电泳以及琼脂糖凝胶电泳进行试验分析。结果试验结果显示EOs对大肠杆菌的最小抑菌浓度(MIC)为5mg·ml-1·在100mg·ml-1浓度的杜香挥发油作用下大肠杆菌的内膜渗透性增大;在浓度为2 MIC的范围内,细菌细胞膜蛋白的荧旋光性发生改变且在一定时间内呈现规律性改变:当杜香挥发油作用浓度逐渐增大(5~0 mg·ml-1),分泌出的蛋白检测含量逐渐增多,细菌细胞膜外膜的通透性逐渐增大,同时细菌DNA的合成含量也发生了变化。讨论:本文初步探讨了天然产物杜香挥发油对革兰氏阴性大肠杆菌的作用机制,初步推测EOs对细菌细胞膜的作用靶标位置可能为细胞膜蛋白,但EOs对细菌DNA的作用尚需进一步研究。

【Abstract】 To investigate the action mechanism of the natural product essential oils(EOs )from Ledum Palustre L.(LP)on Gram-negative bacteria,studies of EOs on the cell membrane,secreted proteins and genomic DNA were carried with Escherichia coli CVCC1418,.UV visible spectrophotometric analysis,fluorescence spectrum scanning,SDS-page electrophoresis and agarose gel electrophoresis.From the results,we know the minimum inhibitory concentration(MIC )of EOs to E.coli CVCC1418 was 5 mg ml-1 and the inner membrane permeability increased at concentration of 100mg ml-1.Otherwise the fluorescence of bacterial cell membrane protein changed and changed regularly in a certain period of time amoung 0~10 mg ml-1. Contents of secretory protein decreased and the outer membrane permeability increased with the increasing concentrations(2.5~0mg ml-1)and the content of the bacterial DNA synthesis changed at the same concentrations. The cell membrane protein may be the target of EOS acting to Gram-negative E.coli and further researches about effects of EOs to DNA still need.LP was a Chinese herb in Changbai Mountain.The essential oils of LP have been utilized for healing different pains,wounds,lung and other diseases in folk medicine.According the research and literatures,LP may be a new resource of functional food factors.
