固定化好氧反硝化细菌及流化床试验
Immobilization of denitrifying bacteria and wastewater treatment in a fluidized-bed bioreactor

【摘要】 利用固定化微生物细胞的流化床反应器进行废水的生物处理,是一个在过去十年间受到持续追捧的新概念。与其他进行废水处理的生物反应器相比,这种流化床反应器具有许多优点。同时,反硝化细菌在硝化过程中的机理及技术特点也激发了人们的兴趣,反硝化细菌在环境生物脱氮过程中具有十分重要的地位。本研究主要针对反硝化细菌106,探讨包埋材料、固化剂、交联时间、固化时间及戊二醛硬化处理等因素对其固定化效果的影响,确定最佳固定化条件,并应用流化床反应器对固定化菌株106进行小型生产试验。实验表明,当以3%海藻酸钠和9%PVA的混合凝胶为包埋材料,以pH6.7的饱和硼酸溶液为固化剂,通过延时包埋法即3%海藻酸钠和9%PVA的混合凝胶交联5h,在固化剂中固定24h制得的凝胶小球性质最好。在此固定化条件下,固定化菌株106的生物脱氮率最高达到了99.81%,与未固定的菌株106的脱氮率相近,凝胶小球的重量膨胀率为0.002g/个,强度为50g/个,同时,在流化床反应器中的使用时间达到了15d。
【Abstract】 Biotreatment of wastewater in a fluidized-bed bioreactor of microbial cells immobilized into particulate solids is a relatively new concept which has gained increased popularity during past decades. The fluidized-bed bioreactor presents many advantages over other bioreactor configurations used in wastewater treatment.The denitrifying bacteria are a group of increasing both fundamental and biotechnological interest and play an important role in the biological nitrogen remove in the environment.In the current study,the effect of several factors,including investment,firming agents,cross-linking time, immobilized time and hardening treatment by glutaraldehyde,on the immobilization of the denitrifying bacteria 106 has been investigated.Based on the optimized immobilizing conditions,the immobilized bacteria 106 has been applied on fluidized bed reactor to determine the denitrifying ability of the immobilized bacteria.The optimal immobilizing conditions were as follow:the gel beads were made through both cross-linking for 5h by the delayed embedding method,the mixed gel made up of 3%sodium alginate and 9%PVA was used as the embedding material and fixed for 24h in the saturated boric acid solution which was taken as the cross-linking agent.Under this condition,the rate of the immobilized denitrifying bacteria 106 reached 99.81%,similar to that of the bacteria without immobilizing.In addition, the weight expansion and intensity of the gel beads was 0.002g and 50g for each separately and the retention time reached 15d in a fluidized bed reactor.
