羟基肉桂酰氨基酸衍生物的抗氧化活性研究

【摘要】 大量的临床试验表明,人体自由基可以引起生物膜脂质和DNA的氧化性损伤,导致癌症、动脉粥样硬化、衰老、脑神经损伤、白内障等多种疾病。咖啡酸,阿魏酸,香豆酸及其酰胺,酯等天然的抗氧化剂在自然界中广泛存在。为了寻找抗氧化活性更好的化合物,以羟基肉桂酸为先导化合物设计合成了一系类羟基肉桂酸衍生物(Structure 1)。研究了这些羟基肉桂酸酰胺类衍生物清除2,2-苯基-1-苦肼基(DPPH)清除自由基的活性,同时以人血红细胞在体外的氧化性溶血为模型研究了自由基AAPH诱导细胞溶血及抗氧化剂对它的抑制作用。结果显示,在两种抗氧化体系中,肉桂酰胺衍生物的抗氧化活性都高于母体化合物咖啡酸、阿魏酸、对羟基肉桂酸的抗氧化活性。
【Abstract】 A large body of clinical and experimental evidences accumulated in the past decade suggested that free radical mediated peroxidation of membrane lipids and oxidative damage of DNA are associated with a variety of chronic health problems,such as cancer, atherosclerosis and ageing.Caffeic acid,ferulic acid,p-coumaric acid and their corresponding amides,esters are natural antioxidants widely present in nature.In order to find potential antioxidants with hydroxycinnamic acid moiety we synthesized a series of hydroxycinnamic acid analogues and compared their antioxidant activities.Their capacity to scavenge 2,2-diphenyl-1-picrylhydrazyl(DPPH) radical and to protect human red blood cells(RBCs) from oxidative haemolysis was used to assess their antioxidant activities.It was found that the caffeic amides exhibited the highest DPPH-scavenging activities while the higher hydrophobic ferulic amides demonstrated the highest antihaemolysis activities.
