泡盛曲霉生产阿魏酸酯酶的研究
Production of Feruloyl Esterases using Three Aspergillus awamori Strains

【摘要】 阿魏酸酯酶能够水解阿魏酸酯中的酯键,将阿魏酸游离出来。本研究以麦麸,蔗渣,玉米皮作为碳源,采用3株泡盛曲霉(分别为40180、40383、2437)进行固体发酵,研究了其阿魏酸酯酶(以去淀粉麦麸作为酶解底物)的产酶活性。结果表明,泡盛曲霉40180的产酶活力最高,最佳培养基组分为:碳源为麦麸与蔗渣(2:1,w/w)、4%(NH4)2SO4、0.233%CuSO4、固液比为1:1,在28℃下发酵3 d,酶活达到最高。
【Abstract】 Feruloyl esterases can break the ester linkage between ferulic acid and the polysaccharides,thus releasing ferulic acid from the plant cell.In this paper,we obtained a strain with high production of feruloyl esterases activity from three strains of Aspergillus awamori (40180,40383,2437,respectively) by solid state fermentation,in which,the enzymic activity was determined by releasing ferulic acid from de-starched wheat bran..It proved that the strain of 40180 has the highest feruloyl esterases activity.And the optimal conditions of enzymatic reaction were determined:temperature 28℃,using wheat bran:bagasse 2:1 as the mix carbon sources,add(NH4))2SO4 and CuSO4 occupy the weight of wheat bran 4%and 0.233%,respectively,base material:water is 1:1,In this condition it can reach the highest peak of feruloyl esterases.
