溶质在皮肤角质层中的分配系数模型研究

【摘要】 农化产品的经皮传质过程已成为农业工程领域中的重要研究热点之一,而农化产品在皮肤中的分配系数是其传质过程的重要参数。目前国内外对亲油性物质在皮肤中的分配系数研究较多,而对亲水性物质分配系数的研究甚少,从而限制了所建立的分配系数模型得适用范围。本研究综合考虑了皮肤、溶质的理化特性对分配过程的影响,对8种亲水性物质分别在皮肤角质层、角质蛋白和脂质域中的分配系数进行测量,并结合已有的分配系数实验结果,建立适用范围更广泛的分配系数预测模型。所建模型比现有模型精度有所提高。
【Abstract】 Agricultural chemicals have been used widely in the agricultural production. While the partition coefficient of the chemicals between the stratum corneum and water was one of the very important parameters in the process of penetration into the skin, it was attracting more and more researchers’ interests. As most of the currently measured experiment values about the partition coefficient using to make the models were about relatively lipophilic compounds, so models have been existed now failed to provide satisfactory prediction for relatively hydrophilic compounds. In this paper, Kscw/w, Kpro/w (partition coefficient of the solute between protein domain and aqueous solution) and Klip/w (partition coefficient of the solute between lipid domain and aqueous solution) of 8 relatively hydrophilic (logKow<0) compounds were measured in equilibration experiments to produce a comprehensive database on partition coefficients. Then the expanded database was used to build a general two-phase partitioning model which can apply not only to lipophilic compounds, but also to relatively hydrophilic compounds. And the model in this study gave a better prediction results.
