麻疯树茎段组织培养及快繁体系的建立

【摘要】 通过比较各种消毒方法和外植体的来源,获得较为理想的芽增殖无菌培养系,为麻疯树离体植株再生奠定了基础。以麻疯树茎段为外植体,配合不同培养基和不同种类、浓度的植物生长调节剂组合进行芽诱导和增殖扩繁、生根诱导以及抗褐化培养。结果表明,腋芽诱导的培养基是以MS基本培养基附加6-BA0.3mg/L为最佳组合。芽增殖培养基的最佳配方为改良MS培养基附加6-BA0.5mg/L。诱导生根的最优基本培养基为MS培养基,且NAA的效果明显优于IBA。Vc是麻疯树抗褐化的最佳选择,其适宜浓度为50mg/L。在本试验所采取的灭菌方法中,取夏季晴天的幼嫩茎段用75%酒精浸泡10s,再用0.1%升汞消毒12min,灭菌效果最佳。

【Abstract】 For further investigations on plantlet production from in vitro stem cultures of Jatropha curc,the stem tissue of Jatropha curca was obtained by comparatively analyzing different disinfection methods and stem sources. When the young stem of Jatropha curca was used as explants,different concentration of growth regulators were added into different minimal mediums to carry out buds inducing and multiplication culture,radication inducing culture and anti-brown culture. The results showed that MS medium in presence of 6-BA 0.3 mg/L was efficiency to induce and germinate buds. The optimum combination of buds multiplication culture was upgraded MS medium combined with 6-BA 0.5 mg/L. The optimum minimal medium of radication inducing was MS medium,and the radication effect of NAA was much better than that of IBA. Vitamin C (Vc) 50 mg/L was in favor of anti-brooding of buds. In order to fix appropriate disinfectants and sterilization time,the explants were firstly marinated in 75 % ethanol for 10 s,then 0.1 % HgCl2 or 2 % NaClO was used for disinfection,and finally,the appropriate disinfectant and the optimum sterilization time were obtained,that is,treating with 75 % ethanol for 10 s,then disinfecting with 0. 1 % HgCl2 for 12 min.
