萧氏松茎象7个地理种群遗传差异的RAPD分析

【摘要】 以萧氏松茎象7个地理种群的基因组DNA为材料,对其进行随机扩增多态性DNA(RAPD)分析,从80个引物中筛选出39个稳定性好、多态性高的引物。7个地理种群共扩增出250条扩增条带,每条引物平均可以得到6.4条扩增条带,其长度从250bp~2000bp不等,其中200条扩增条带为多态性条带。通过公式计算多态性位点、遗传距离和遗传相似性,结果表明:萧氏松茎象7个地理种群之间的遗传距离指数的变异范围为0.2433870~0.502646,遗传相似性指数为0.497354~0.756613,平均遗传距离指数为0.388763。其中,福建宁化与广西临桂地理种群间的遗传距离最小,为0.243387,两者之间亲缘关系最近。而广东英德和江西永修地理种群间的遗传距离最大,为0.502646,表明这两个种群间有较大的遗传背景差异。

【Abstract】 Seven geographical populations of Hylobitelus xiaoi Zhang are detected by RAPD. Thirty-nine primers are screened from 80 random primers and amplified 250 bands(from 250bp to 2000bp). 200 of theses bands are polymorphic bands. The average number of obtaining segments of individual primer is 6.4. Polymorphisms, genetic distance and genetic similarity indices were calculated. The results of this study showed, The genetic comparability of seven geographical populations varies 0.497354~0.756613 and the range of genetic distance is from 0.2433870~0.502646 and the average was 0.388763. The genetic distance between the two populations from Ninghua,Fujian and Lingui,Guangxi is the smallest among seven populations. However, the genetic distance between the two populations from Yingde,Guangdong and Yongxiu,Jiangxi is the biggest.
