赣南油茶高产优良无性系的ISSR分子鉴别

【摘要】 采用ISSR分子标记方法,对赣州市林业科学研究所培育的33个高产油茶优良无性系进行分子鉴别研究。采用优化的ISSR-PCR体系,从100条ISSR随机引物中筛选出18条多态性高、条带清晰的引物进行PCR扩增,共得到202个位点,其中182个为多态位点,多态比率达90.1%;建立了33个油茶无性系的DNA指纹库,较准确地进行了各优良无性系的分子鉴别;聚类分析结果表明,各无性系遗传背景复杂。本研究结果可为油茶新品种的登记和保护及分子标记辅助选择育种提供分子依据。
【Abstract】 ISSR were used to detect the genetic diversity among 33 oil-tea superior clones in southern jiangxi, which were bred by forestry science institute of Ganzhou.18 specific primers were screened out from 100 ISSR random primers, and 202 bands were amplified from 33 clones with 182 polymorphic loci (ratio 90.1%). The result showed that there was a big genetic distance between them and the DNA fingerprint of oil tea superior clones was established, providing a molecular base for the protection of new varieties and molecular breeding of oil-tea.
