光皮树无性系的ISSR分子标记

【摘要】 应用ISSR技术对光皮树的16份材料(包括湖南省林业科学院选育出来的15个无性系和1个普通光皮树对照)进行分子鉴定,从28个引物中筛选出10个ISSR引物,扩增得到106个扩增点,大小分布100-2000bp之间,多态性位点为90个,多态性比率为84.9%。利用Popgene3.2、DPS8.0和NTsys2.10e软件对ISSR扩增结果分别进行多态性分析、聚类分析和主成分分析,结果表明:ISSR具有较高的多态性,平均每个位点观察等位基因数为1.84,每个位点有效等位基因数为1.403,Nei多样性指数为0.2878,Shannon信息多样性指数为0.4472;ISSR具有较高的特异性,将光皮树聚为四类;ISSR具有鉴定的高效性,只需AG91515和AG91503引物就可为16份光皮树材料建立ISSR鉴定识别卡。
【Abstract】 Inter-simple sequence repeat (ISSR) analysis was conducted to identify 16 Swida wilsoniana samples (including 15 clones selected by Hunan Forestry Academy and 1 local variety). Of the 106 discernible fragments with a ranged from 100bP-2000bP by 10 primers selected (among 100 primers), 90 appeared to be polymorphic. The percentage of polymorphic bands (PPB) was 84.9%. The ANVOA analysis, UPGMA analysis and PCA analysis of the ISSR amplification result were studied with the Popgene3.2, DPS8.0 and NTsys2.10e software, respectively. The result showed: ISSR marker had high polymorphic in 16 Swida wilsoniana samples, with the mean number of observed alleles (Ao) 1.84, the mean number of effective alleles (Ae) 1.403, genetic diversity (He) 0.2878 and Shannon information index (Ho) 0.3191. 16 Swida wilsoniana samples were divided into four groups with ISSR marker as an effective and specific method for cultivar identification, whose ISSR identification cards were done base on the amplification results of two primers AG91515 and AG91503.
