【摘要】 目的:联合检测Pokemon蛋白及P53蛋白在胃上部癌组织中的表达,判断外科手术切缘相对安全的范围;同时为以后胃癌患者在外科治疗方面提供理论依据。方法:收集2011年12月到2012年3月哈尔滨医科大学附属第三医院胃肠外科的60例胃癌患者行根治性手术切除的肿瘤组织。将每个患者的肿瘤组织的取材部位分为对照组和胃癌组,分别对应胃癌的中心部位和肿瘤边缘的1.0cm,1.5cm,2.0cm,2.5cm,3.0cm3.5cm,4.0cm,4.5cm,5.0cm处的癌组织,在以上每个实验点去取材(大小为0.5cmx0.5cm),然后采用Western blot检测每处Pokemon蛋白,P53蛋白的表达并进行蛋白表达量的比较,最后统计数据,运用SPSS统计软件进行系统分析。结果:胃癌组织中Pokemon蛋白与P53蛋白高表达,明显高于其他癌旁组织,其中在肿瘤中心的蛋白表达量Pokemon蛋白高达63.3%(38/60),P53蛋白占55%(33/60);对照组与胃癌组蛋白表达的关系大致可描述为并随着距离的增加相应的减少,即阳性率趋向于零;在胃上部癌中P53,Pokemon蛋白的表达同时在2.5cm处出现统计学的差异(P<0.003),在3.5cm及以上组织中出现表达率为阴性。结论:1.Pokemon蛋白和P53蛋白在胃癌组织中高表示,有可能在胃癌的发生发展及演变过程中充当主要角色;2.Pokemon蛋白和P53蛋白肿瘤重要标记物,可根据着两种蛋白的表达作为临床治疗的评估指标;3.以Pokemon,P53蛋白的表达作为肿瘤标记物,发现3.5cm及以外的组织中此两种蛋白的表达与正常组织无差异,对外科手术切缘距离的确定可能具有一定指导意义。更多还原

【Abstract】 Objective:To investigate the expression of Pokemon and p53 in Gastric carcinoma and their clinical significance and To investigate the safe distance of surgical margin by the molecular margin represented by Pokemon and P53,which can provide basis for surgical treatment Methods:In 60 cases of gastric cancer.,specimens were get from tumors’ tissue and every 0.5 cm from the proximal tumor margin at the beginning of the 1.0 cm to distal.using the Western-Blot to find Pokemon and P53 expression,and analysing statistical significance.Results High expression of Pokemon,P53 in the tumors’ cental tissue.The farther distance of tumor margin,the lower expression of Pokemon protein.The amount of protein expression in cancer center of Pokemon protein up to 63.3%(38/60),P53 protein was 55%(33/60);According to the expression of the Pokemon、P53proteins,there were significant different at the 2.5cm(P<0.003).the positive expression rate of Pokemon、 P53 proteins were 0 at 3.5cm.Conclusion:1.Expression of Pokemon protein and P53 protein in stomach carcinoma group is high,said may be a major role in the process of in the stomach cancer development.2.Pokemon,P53 proteins are important tumor markers,according to the expression of the two proteins as indicators of clinical assessment.3.The using of the positive expression of the Pokemon、 P53proteins as a basis of the surgical margin of the gastric cancer showed that 3.5cm was a safe distance,which may have guiding to the surgical margin.更多还原
