Rh/Cu串联催化合成6-酰基吡啶[2,1-a]并异吲哚
Facile Synthesis of 6-Acylated Pyrido[2,1-a]isoindoles via Rh/Cu-Cocatalyzed Tandem Reactions

【摘要】 基于过渡金属催化的C-H键活化构筑有机化合物骨架长期以来一直备受关注[1,2],特别是Rh(III)/Cu(II)催化体系的研究在近年来取得了显著的进展[2]。本工作发展了[RhCl(COD)]2/Cu(OAc)2.H2O共催化下,2-芳基吡啶衍生物与γ-取代炔丙醇反应,经过炔基化中间体A,高效合成C-6位酰基取代的吡啶[2,1-a]并异吲哚的新方法 (Fig.1)。该反应也可拓展到由2-乙烯基吡啶衍生物与γ-取代炔丙醇反应合成C-3位酰基取代中氮茚。
【Abstract】 Construction of various frameworks of organic compounds via transition metal-catalyzed C-H bond activation has continuously attracted great attentions.[1,2] In particular, impressive progress has been made for the Rh(III)/Cu(II) catalyst system in recent years.[2] We herein reported the [RhCl(COD)]2/Cu(OAc)2.H2O-cocatalyzed tandem reactions for the facile synthesis of 6-acylated pyrido[2,1-a]isoindoles from 2-aryl pyridine and 1,3-diphenylprop-2-yn-1-ol via the alkynylated intermediate A. This reaction can be extended to synthesize 3-acylated indolizine derivatives from 2-vinyl pyridine derivatives and 1,3-diphenylprop-2-yn-1-ol.
