Pb(PPh34催化Suzuki偶联反应合成3-芳基-2,5-二甲基吡嗪衍生物

【摘要】 2,5-二甲基吡嗪属于吡嗪类香料中的一种,可作为香味添加剂少量添加到食物中,除此之外它还是合成吡嗪类药物的重要活性中间体[1]。吡嗪杂环及其衍生物可通过取代苯硼酸和卤代物在Pd催化下,应用Suzuki偶联反应合成,该合成方法操作简单且条件温和;原料氯代芳烃廉价易得,有利于工业化生产;并且允许芳烃上有多种官能团,因此非常适合制备各种官能团取代的吡嗪衍生物[2]。本文以2,5-二甲基-3-氯吡嗪为起始原料,利用Suzuki偶联反应来构筑分子骨架,合成了几个不同取代基修饰的新型吡嗪衍生物。这些化合物结构经1H NMR,IR,EI-MS分析表征。结果表明:在无氧条件下的碱性二氧六环中,使用Pd(PPh3)4做催化剂,所有反应均可有效的进行,除邻氰基苯硼酸产率较低外,其它产物的收率均较高,其中多数产物没有作为药物活性基团被报道过。

【Abstract】 Pyrazines are important components of aroma fragrances and have been recognized as flavor components in foods. Pyrazine derivatives are known for used as potential pharmacophore of a large number of biologically active substances. Because of the wide variety of applications associated with the pyrazine moieties, their synthesis has remained the goal of many research groups over the years. Among the various methods developed, pyrazine compounds are synthesized by the Suzuki coupling reaction in presence of Pb(PPh3)4. We report herein the protocol comprises a direct coupling between the 2,5-dimethyl-3-chloropyrazine with substituted phenylboronic acid in basic aqueous dioxane. We attempted our procedure using a number of both structurally and chemically diversified phenylboronic acid to synthesize pyrazine derivatives(See fig. 1) and were able to get desired results in each case except o-cyanophenylboronic acid. Most of these products have never been reported in literatures as potential biologically active substances.
