DNA/AgNPs双光子纳米探针的构建及细胞GSH成像研究

【摘要】 谷胱甘肽(GSH)是生物体内重要的抗氧化剂和自由基清除剂,对于维持生物体内氧化还原平衡具有重要意义。本文中,我们构建了DNA/AgNPs双光子纳米探针,并用于GSH检测及细胞成像。该探针基于双链DNA与双光子染料BMVC-Bu的相互作用和荧光增强效果及DNA模板合成AgNPs对BMVC-Bu的荧光猝灭作用,目标物GSH与AgNPs的作用可以解除AgNPs对BMVC-Bu的猝灭作用,从而实现对GSH的检测。利用BMVC-Bu的双光子荧光信号强度的变化,可以有效避免生物体内自体荧光的干扰。该探针对GSH响应灵敏,可以实现低浓度GSH的检测,在0.1–15 uM GSH浓度范围内具有良好的线性关系。

【Abstract】 GSH, as a oxidation inhibitor and cleaner of free radical, plays a key role in maintain the ReDox balance in biological system. We have developed herein a two-photon DNA/AgNPs nanosensor for GSH detection and imaging. We have synthesized a two-photon dye BMVC-Bu, which can interact with duplex DNA following great fluorescence increase. The nanosensor is composed of DNA template AgNPs and the complex of BMVC-Bu/DNA. The sensing approach is based on the specific interactions between GSH and DNA-templated AgNPs. In the presence of GSH, the fluorescence of BMVC-Bu would be restored as a result of the robust Ag-S between AgNPs and GSH. Due to the bright two photon-excited fluorescence of BMVC-Bu, a low concentration of GSH can be determined even in a complex system, showing a good linear relationship in the range of 0.1 – 15 μM.
