肿瘤组织非靶向质谱分子成像新方法研究

【摘要】 目的:建立基于空气动力辅助离子化技术的非靶向质谱分子成像新方法(AFAI-MSI),并用于恶性肿瘤组织的分子筛查;方法:采用AFAI-MSI方法,获取肿瘤组织中内源性小分子的分布特征、采用目标区域数据提取和多变量统计分析,发现与恶性肿瘤相关的潜在生物标志物;结果:通过系统关键参数的优化,建立了非靶向AFAI-MSI成像方法,获得脂质类等内源性小分子在肿瘤组织中的分布特征;结论:AFAI-MSI方法可从分子水平上识别肿瘤组织与正常组织,有望为恶性肿瘤的临床诊断和分子病理研究提供一种新的分析工具。

【Abstract】 Objective:To develop an untargeted molecular imaging method for the detection and delineation of human cancerous tissue using air flow assisted ionization-mass spectrometry(AFAI-MS).Methods:The AFAI-MSI method were adopted to acquire mass spectra along with position information of endogenous biomolecules,data extraction from region of interest,multivariate statistical analysis and visualization were carried out to discover and validate tumor biomarkers.Results:AFAI-MSI method has been established through the optimization of its key parameters and has been successful to trace the spatial distributions of hundreds of endogenous biomolecules in tumor tissue.Conclusion:AFAI-MSI is a new powerful analytical technique with the capacity to discrimination tumor and normal tissue by imaging the distribution of biomolecules,which has the potential to emerge as a new tool for the cancer diagnosis and molecular pathology study.
