适体修饰纳米金共振瑞利散射光谱法检测痕量妥布霉素

【摘要】 用硼氢化钠还原氯金酸制备了金纳米粒子(GN);用适配体(Apt)修饰NG可获得较稳定的Apt-GN探针。在pH6.8PBS缓冲液中及NaCl存在下,Apt-GN探针稳定而不聚集。当有氨基糖苷类抗生素-妥布霉素存在时,妥布霉素与Apt-GN探针中的Apt特异性结合并释放出纳米金。纳米金在NaCl作用下聚集,导致体系在368nm处的共振瑞利散射光强度增强。其共振散射峰峰强度的增大值△I与妥布霉素浓度在6.7-66.7ng/mL范围内呈现良好线性关系。该法用于分析测定妥布霉素滴眼液中的妥布霉素,结果令人满意。
【Abstract】 Gold nanoparticle(GN) was prepared by the sodium borohydride-reduced procedure,that was modified by the aptamer(Apt) to obtain stable Apt-GN probe for tobramycin.In pH 6.8 PBS buffer solutions and in the presence of NaCl,the Apt-GN probe was stable.Upon addition of tobramycin,it reacted with the Apt in the probe to form very stable complex and to release GNs.Under the action of NaCl,the GNs were aggregated to big cluster that caused the resonance Rayleigh scattering(RRS) peak enhanced at 368nm.The enhanced intensity ΔI was linear to tobramycin concentration in the range of 6.7-66.7 ng/mL.This Apt-GN RRS method was applied to analysis of tobramycin in real sample,with satisfactory results.
