基于磁性石墨纳米晶的生物检测应用

【摘要】 因为其独特的物理和化学性质,磁纳米晶受到了人们越来越多的关注并在许多领域都有广泛的应用,特别是在生物应用领域,是大家关注的热点。碳纳米材料,作为一种新型先进功能材料也是研究的热点。在磁纳米晶表面包裹上一层无机碳层将能有效地提高磁纳米晶的各种性能。我们利用化学气相沉积(CVD)的方法制备了新型石墨包裹的磁纳米晶颗粒。通过表面聚合物修饰的方法,我们对表面包裹的石墨层进行表面改性,石墨包裹的磁纳米晶的水溶性、稳定性和生物相容性得到了提高,并有效的提高了抗非特异性干扰的能力。改性后的磁纳米晶被用来进行细胞的染色、操控、富集以及检测。磁纳米晶染色的细胞能在外部磁场的调控下可控的定向运动。除此之外,磁纳米晶还被制成点阵微芯片用来富集和检测全血样品中的微量癌细胞。从数百个到只有两个混到1毫升全血溶液中的癌细胞都可以高效的被磁纳米晶点阵芯片检测出来,具有广泛的生物医学理论意义和临床实用价值。我们还通过设计多功能、程序可控的DNA捕获、富集与检测策略,开发了基于磁性石墨纳米晶的生物检测平台体。平台对DNA分子的检测限达到了50pM以下,对比常规基于石墨烯或者碳纳米管等碳材料的荧光生物检测提高了3个数量级,显示了很高的检测灵敏度。在检测选择性方面,我们也考察了MGN传感平台对碱基错配的区分能力,也体现了非常好的检测特异性。我们这种检测方法,简单,便宜而且不需要生物酶的参与就能得到非常高的检测能力,为生物医学特别是分子医学的诊断提供非常好的工具。
【Abstract】 Graphene is a two-dimensional atomic crystal which consists of single-layer carbon atoms densely packed into a honeycomb lattice. Owing to its unique physical, mechanical, and electronic properties, graphene has attracted considerable interest as a desire material for different applications. Meanwhile, magnetic materials, especially magnetic nanoparticles, are of particular significance, and a wide variety of applications have been envisaged for them. We synthesized a core-shell magnetic graphitic nanocapsule (MGN) material and investigated its capabilities for the detection of biomolecules and cells. We designed a multipurpose DNAcapturing and releasing strategy for programmed and targeted fishing, enrichment and detection of DNA. This MGN platform exhibited good sensing specificity and selectivity for low concentration DNAsolution. Cell staining, manipulation, enrichment and detection were also developed with these MGNs. Cancer cells from hundreds down to only two were able to be simply and efficiently detected from 1 mL whole blood in a microarray fashion.
