【Abstract】 In the HPLC-MS top-down strategy,the mixed proteins are separated first by HPLC with no need for pre-proteolysis.Because the number of proteins are much less than the correspond traptic-peptides,the HPLC separation can be nice done even with baseline resultion for two conjunctive proteins.In the previous study,we have invented a on target-digestion technique,in which the protein effulent was dipped into MALDI target plate with an pre-immobilized-trapsin.The depped protein can be digested within a minute and then ready for MALDI-TOFMS identification.In addition,we have developed the other on-line proteolysis technique.In this techinique,the protein effulent is introduced into a segment-bobble columm,where the protein solution is stayed in bobble in vivo mixed with trypsin sultion and nitrogen gas was segmented bobble.更多还原
