高速逆流色谱法分离制备槐花中的黄酮类化合物

【Abstract】 Two flavones were prepared,isolated and purified from Sophora japonica by high-speed countercurrent chromatography(HSCCC) for the first time using ionic liquids to improve the separation effect.A two-phase solvent system composed of n-hexane-ethylacetate-ethanol-water-acetic acid and ionic liquid was used.The obtained fractions were analyzed by HPLC to test its purity and to ensure the compound.The results indicated that 18.2 mg of rutin and 9.6 mg of quercetin with their purities over 97 % were obtained from 50 mg crude extract of Sophora japonica.By adding ionic liquid,the retention time of the compound decreased from 80 min to 50 min,and the resolution increased from 0.9 to 1.8.The separation effect was significantly improved and complete separation was achieved.The established method can be used for the separation and purification of flavones from the Sophora japonica.
