基于Staudinger反应的ATP引发的三明治结构荧光探针

【Abstract】 Analyte addition and catalyze the organic reaction which leading to a fluorescence product represent an attractive strategy for the detection of biomolecules.The Staudinger reaction is between a phosphine and an azide and give a primary amine and the corresponding phosphine as product.The reaction is biocompatible and orthogonal to most biological functionalities.We synthesize a 7-azidocoumarin whose fluorescence is quenched by introduce an azide group.Upon addition of triphenylphosphine(TPP),the Staudinger reaction take place and fluorescence recover on mM concentration.We report the application based on an ATP assisted sandwich panel structure which draw the distance between two divided ATP aptamer coujugated with 7-azidocoumarin and TPP,then catalyze the Staudinger reaction under lower concentration and get fluorescent 7-aminocoumarin as signal product for the detection of ATP.
