siRNA/壳聚糖纳米粒的制备和分析

【摘要】 采用复凝聚法制备载siRNA的壳聚糖纳米粒,用琼脂糖凝胶电泳分析质粒的纯度和纳米粒的电性结合,TEM观察纳米粒的粒径和形态,并计算该纳米粒的包封率。结果标明:采用复凝聚制备法能够实现壳聚糖对的包埋,壳聚糖与siRNA电性结合稳定,包封率达到92.8%,制备的壳聚糖质粒纳米粒呈亚球型,粒径为50~100nm。
【Abstract】 SiRNA was loaded into chitosan nanoparticles by using the complex coacervation method.The purity quotient of the plasmid and the integration of the nanoparticles were analyzed by agarose gel electrophoresis,the grain diameter and shape of the nanoparticles was determined by TEM.The encapsulation ratio was also calculated.The results showed that the chitosan and siRNA was integrated stably by electrostatic interaction.The encapsulation ratio of nanoparticles was 92.8%The nanoparticles were nearly spherical in shape with the diameter of 50~100 nm.
