叶酸偶联壳聚糖的分离分析

【摘要】 采用葡聚糖凝胶柱分离纯化自制的叶酸偶联壳聚糖,用紫外分光光度法监测分离情况,并借助紫外分光仪,测定了叶酸偶联程度,使用红外光谱对偶联产物进行分析。通过葡聚糖凝胶柱可成功分离叶酸偶联壳聚糖与未反应的叶酸活性酯,红外光谱分析进一步确认了叶酸被成功地接枝到壳聚糖上,此叶酸偶联壳聚糖有望为肿瘤靶向治疗提供良好的基因递送载体。
【Abstract】 The self-made folate-conjugated chitosan(FA-CS) was separated via sephadex G-10 column.The structure and the coupling ratio of FA-CS were investigated by infrared(IR) spectrum and ultraviolet(UV) spectrophotometer. The results showed that FA-CS could be completely separate from unreacted activated folic easter by Sephadex G-10 column.The product was conjugated successfully between folate acid and chitosan.Thus the self-made FA-CS is expected to be a potential new gene carrier for tumor cell-selective targeting.
