钯(Ⅱ)离子与巯嘌呤相互作用的共振瑞利散射光谱及其分析应用

【摘要】 在pH 4.8的BR缓冲溶液中,金属离子Pd2+与巯嘌呤反应形成稳定配合物,其物质的量之比为1:1,同时引起共振瑞利散射(RRS)的显著增强,并出现新的RRS光谱。Pd2+与药物的反应产物具有较宽的散射峰,选取400 nm处为测定波长。在一定范围内散射增强(ΔI)与药物的浓度成正比,线性范围是0.05~3.6μg/mL。方法具有较高的灵敏度,检出限(3σ)为17.0 ng/mL。据此建立的分析方法有较好的选择性,可用于血清、尿样中巯嘌呤的测定,能获得较满意的结果。

【Abstract】 In pH 4.8 B-R buffer solution,The metal ion Pd(Ⅱ) could react with Mercaptopurine(MP) to form stable coordination compound,theirs moles ratio was 1:1.At the same time the resonance rayleigh scattering(RRS) spectra were been enhancemented and appeared new and wide RRS peak,we selected the highest peak located at 400 nm as detection wavelength.Because there was a linear relationship between the RRS enhanced intensity(AI) and the MP concentration,and the linear range were 0.05~3.6μg/mL and detection limit(3σ) was 17.0 ng/mL,So a new method for determination of the MP can be developed.The method had fine selectivity and had been applied to determination of the MP in serum and urine sample with satisfaction result.
